Validation of a human saliva model for the determination of leachable monomers and other chemicals from dental materials

 

Although saliva and different types of artificial saliva have been used as media to study the leakage of chemicals from dental resin materials, reported studies often do not demonstrate the validity of replacing saliva with artificial saliva. In this study, we aimed to directly compare the performance of artificial saliva with that of unstimulated saliva. To this end, we developed and validated an LC-MS/MS method for the quantification of selected common leachables in unstimulated and artificial saliva. The study showed that artificial saliva can be a suitable replacement for native saliva, but might underestimate leakage of more lipophilic methacrylates such as BisEMA.

 

The artificial saliva used in this study contained nine compounds including salts, small organic molecules, mucin, and amylase. Unstimulated saliva was collected from employees at NIOM. We selected a simple strategy for sample preparation in which the target chemicals were extracted with methanol. During method validation, we compared the concentrations of selected leachables from spiked saliva samples. A leaching trial was also performed using 3D-printed materials.

 

The performance characteristics of the method were comparable for unstimulated and artificial saliva samples. The lowest lower limit of quantification (LLOQ) achieved was 1.5 ng/mL, while the highest LLOQ was 90 ng/mL. The combined measurement uncertainty for individual chemicals at a concentration of 10 times the LLOQ was less than 20% for both unstimulated and artificial saliva, with the exception of the methacrylate BisEMA. In the leaching trial using 3D-printed materials, the release of BisEMA was higher in unstimulated saliva than in artificial saliva.

A week after officially ending the Network Conference 2024, all that is left to say is “Thank you” to all of those who joined us!

 

We are still energised from being surrounded by more than 140 competent people from all over Norway who where gathered to learn more about research and innovation in oral health.

 

Two days filled with exciting topics in dental research, success stories, career development, research funding, and theme groups. New relationships were made, networks were nurtured, and a foundation for future collaboration was laid.
